細(xì)胞核膜是包裹著真核細(xì)胞核、分隔細(xì)胞核和細(xì)胞質(zhì)的雙層膜結(jié)構(gòu);核孔復(fù)合體則是鑲嵌在內(nèi)外核膜上的蛋白復(fù)合體,主要由胞質(zhì)環(huán)、核質(zhì)環(huán)、核籃等亞結(jié)構(gòu)組成;核膜蛋白包括核孔復(fù)合體的功能十分重要,其蛋白突變會誘發(fā)人類多種疾病,比如早衰癥、阿爾茨海默病、病毒感染、癌癥誘發(fā)等等。目前在植物中僅發(fā)現(xiàn)少數(shù)幾種核膜蛋白,它們參與植物的生長發(fā)育和不同環(huán)境適應(yīng)中。植物核膜蛋白和核孔復(fù)合體的組成和功能仍需要深入研究。本課題組以擬南芥、番茄和小麥等模式植物,利用細(xì)胞生物學(xué)、分子生物學(xué)、遺傳學(xué)、蛋白結(jié)構(gòu)解析、生物信息學(xué)并結(jié)合多組學(xué)等方法,在前期研究結(jié)果的基礎(chǔ)上繼續(xù)探索植物核膜蛋白,包括挖掘新的內(nèi)外核膜蛋白和核孔復(fù)合體組分,構(gòu)建其中的互作網(wǎng)絡(luò)圖譜,深入研究它們的生理功能和調(diào)控機(jī)制,并應(yīng)用于改良農(nóng)作物性狀、培育高產(chǎn)抗逆作物新品種。

(1)通過建立兩種蛋白組學(xué),挖掘了一批新的植物核膜蛋白
(2)揭示了植物核膜蛋白與染色體互作的新機(jī)制
